Contraindications

Acute intermittent porphyria

Barbiturates stimulate synthesis of aminolevulinic acid synthetase, which is the rate limiting enzyme in the formation of porphyrin.

In AIP, defective porphobilinogen deaminase causes buildup of porphobilinogen.
