Absolute contraindications
Acute intermittent porphyria: Barbiturates stimulate synthesis of aminolevulinic acid synthetase, which is the rate limiting enzyme in the formation of porphyrin. In AIP, defective porphobilinogen deaminase causes buildup of porphobilinogen.
